Inhibition of 3-hydroxy-3-methyl-glutaryl-coenzyme A reductase enzyme by dipeptides identified in dry-cured ham

Abstract High cholesterolemia is a key risk factor for the development of cardiovascular diseases, which are main cause mortality in developed countries. Most therapies focused on modulation its biosynthesis through 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMG-CoAR) inhibitors. In this sense, food-derived bioactive peptides might act as promising health alternatives their ability to interact with crucial enzymes involved metabolic pathways, avoiding adverse effects synthetic drugs. Dry-cured ham has been widely described an important source naturally-generated exerting ACEI-inhibitory activity, antioxidant and anti-inflammatory activity between others. Based these findings, aim work was assess, first time, vitro inhibitory HMG-CoAR exerted by dipeptides generated during manufacturing dry-cured ham, previously relevant roles other bioactivities. The assessed measuring substrate consumption rate 3-hydroxy-3-methylglutaryl CoA presence, following pertinent calculations. Further research carried out estimate possible interactions most enzyme performing silico analysis consisting molecular docking approaches. Main findings showed DA, DD, EE, ES, LL Additionally, computational indicated statin-like HMG-CoAR. This study reveals, hypocholesterolemic potential ham-derived and, at same converges vein many reports that experimentally argue benefits due peptide content.
